In short - no 3rd brother in Conan Doyle's original stories. Then, William S. Baring-Gould wrote a fictional biography of Sherlock where Sherrinford, a 3rd brother was introduced. In the series, they made a twist and changed the brother to a sister, while playing around with the name Sherrinford (in Mycrofts notebook) which you'll find out what it is in the final episode.
