the hundred knights are just the number of troops that Benimaru is eliminating. they aren't special, and it has nothing to do with who killed the people of the town.

The troops that guarding the totems + people in the west were the one who committed massacre in the town. Now they are waiting at the totem for reinforcement to come while protecting it. The others who are not involved was the 20,000 army who is coming, which rimuru is heading.

Well the one that lead the raid on Tempest, Folgen, is currently with the army of 20.000 and those protecting the totems seem to be wearing the Temple Knight's armor/uniform and not that of the Falmuth army, which makes sense considering what they are protecting was set up by Archbishop Reyhiem. So it seems more like, while the Temple Knights set up the barrier, the Otherworlders were sent into Tempest to cause a commotion, and once the Barrier is up Folgen and 100 Knights/cavalry men rush in to attack but then retreat back the the rest of Falmuth army while the Temple Knights and Otherworlders keep the barrier protected and make sure no one escapes.

This is something the Anime skips ... and I do think they could have included it instead repeating the the whole story and maybe even shortening the flashback sequence ... and at least mention this The reason the Otherwolders are positioned to the west is because Falmuth expects the humans that were in Tempest to try and escape into the friendly nation of Blumund to the west so their plan is to kill everyone fleeing to the west including fellow humans since witnesses that could tell what actually went down in the town would be an inconvenience. As such Rimuru basically opens a portal that allows a small number of people to go through at a time, since he is not able to mass teleport them ...

Yep, I was intending on saying the same. And all the human civilians have now been evacuated, unbeknownst to the Falmuth army. There is a tendency in the anime to skip what I at least think is important information.

The composition of the main army is not exactly what you think it is. Also I think your take on Rimuru's insistence on doing this himself is more about reminding himself not to get complacent than trying to protect his people from having to see him doing it, imo. Monsters are by nature pretty survival of the fittest, strength is everything types, so I don't think he would shock them. So he actually lied when he said he had to do the killing himself in order to collect the souls. All that is necessary is for his people do it on his behalf. (It might also have to be people connected to him through naming. One of the things the anime glosses over is the importance of these connections. They pass skills and strength to each other in a positive feedback loop, with Rimuru as the nexus, making each of them stronger. Naming is one of the secrets to Rimuru's strength. No one else does it to the extent Rimuru does.). Oh, and now Rimuru suspects Clayman of not having become a True Demon Lord. There are ways of being recognized as a Demon Lord without being more than a Demon Lord Seed; I won't go into how that can happen here.

The last things Rimuru says, to me are him putting himself into the proper mindset for what he's planning to do. Btw, the word Rimuru uses for food here, esa, means animal feed, fodder. He is dehumanizing them in his mind purposefully. Also, I don't think it's just the energy of the souls by themselves that cause the evolution. It's not entirely clear to me what exactly happens, but for one thing, everybody has a soul, but only human souls have this special property. The souls have to be basically intact when Rimuru takes them in. Back in the town, Rimuru has done everything he could to preserve both the souls of the dead monsters and their bodies. He cast preservation spells on the bodies and, and one of the things he does to keep the souls intact is to increase the concentration of magicules in the air within the barrier as a positive pressure measure against dissipation.
